Unusual mitochondrial ribosome from a parasite

Complexes of proteins and RNA known as ribosomes play the crucial task of translating genetic messages, in the form of mRNAs, into proteins. Ubiquitous in eubacterial, eukaryotic cells, in the archaea, and in organelles, these workhorses of the genetic information system differ markedly in composition among the phylogenetic domains. Manjuli Sharma et al. found that the mitochondrial ribosome of the Leishmania tarentolae (Lmr) parasite contains several features previously unseen in cytoplasmic or organellar ribosomes. The Leishmania parasites, which are transmitted by the bite of sand flies, cause skin sores or potentially fatal visceral disease. The authors obtained a 3D, cryo-electron microscopic map of the Lmr parasite and discovered that, in addition to having lower RNA content, this mitochondrial ribosome was more porous than other known ribosome structures. Major portions of key functionally conserved sites, such as the channel where mRNA enters, the transfer RNA passage, and the region where nascent polypeptides exit, contain Lmr-specific proteins, rather than familiar ones. Despite the specificity in these regions, the overall architecture of the Lmr has been conserved, the authors say, suggesting the imposition of tight constraints on this evolutionarily divergent ribosomal form.
